Dustin Hoffman seriously wanted to play the role of Dewey Wilson, but was refused the part by director Michael Wadleigh who really wanted to work with Albert Finney, who was his favorite actor. It was the only time in his career that Hoffman was rejected for a part.
Wolfen ending explained (spoilers)

Dewey (Albert Finney) and Rebecca run away from the wolfen (they look just like ordinary wolves but they're not.. it's suggested that they're spirits or gods) and go to van der Veer's apartment. Dewey smashes the scale model of the van der Veer building as he and Rebecca are surrounded by the wolfen. When Dewey smashes the scale model, the wolfen understand that their territory won't be turned into buildings and parks (as indicated by the model). The wolfen disappear. The last scene is Eddie (Edward James Olmos) and his friends looking at the city from a bridge...
