This was the least financially successful entry in the "Scary Movie" series.

The saying "Klaatu Barada Nikto" was also used in The Evil Dead (1981) by Ash, to summon up the demon from the Necronomicon Ex Mortis, the Book of the Dead. (Ash also screws up reciting the phrase, thus causing all sorts of problems in Army of Darkness (1992) too). Incidentally, Nikto was also the name of a character that appeared in Jabba the Hutt's palace in Star Wars: Episode VI - Return of the Jedi (1983). The saying originates from The Day the Earth Stood Still (1951) and is used to 'calm' Gort down.

Director Malcolm D. Lee did not like this film. He stated that Bob Weinstein called him to direct, told him there were requirements, and stated that the producer constantly interfered with this film.

Marisa Saks, who plays Amy, is the great niece of Leslie Nielsen. Nielsen played the President of the United States in Scary Movie 3 (2003).

Marlon Wayans, who originally created the "Scary Movie" franchise alongside his brothers Keenen Ivory Wayans and Shawn Wayans back in 2000, said in an interview that the studio asked the Wayans Brothers to return for Scary Movie 5 (2013), but the brothers declined. Marlon Wayans later released his own horror movie spoof, A Haunted House (2013), which performed so well at the box office that it spawned a sequel, with A Haunted House 2 (2014) released the following year.

Ted appeared on the cover but did not make the film's theatrical final cut. In the Blu-ray extended version, Ted appears in the Jody Black Swan intimate, drug fueled encounter with Kendra, as among the many bedroom attendees in intercourse.

Unlike the previous "Scary Movie" films, this is the only one that has a "happy ending," with the only actual deaths being Charlie Sheen's, the teenagers at the cabin, and Mama's.

Even though a majority of scenes were deleted from the movie, the DVD and Blu-Ray only included 8 of the deleted scenes. The first trailer showed snippets of other deleted scenes that were not included. Audiences were curious if there was an original cut without the reshoots. That has never been released.

David Zucker directed the additional/re-shoots from January to February 2013. Main actress Ashley Tisdale said in several interviews that the material from the re-shoots (and ADR sessions) made about 60% of the final theatrical cut. Director Malcolm D. Lee did not have anything to do with the editings since he was already engaged in doing The Best Man Holiday (2013) by the time they did them. Lee did not even show up at the Scary Movie 5 (2013) premiere in Los Angeles, for unknown reasons.

Anna Faris did not return for Scary Movie 5 (2013) due to her pregnancy. Other cast members from the previous films, such as Regina Hall, Kevin Hart and Anthony Anderson, were offered roles but opted not to return.

The Hunger Games (2012) was spoofed in a deleted scene, which was supposed to be included on the DVD/Blu-ray.

Jerry O'Connell also starred in Scream 2 (1997). The first Scary Movie spoofed the first two Scream films among other horror films.

Since Here Comes Honey Boo Boo (2012) was not very known in Germany, the reference to the show was replaced by I'm a Celebrity, Get Me Out of Here! (2003), a hit reality series on German TV, in the German-dubbed version. Similar changes were also made in other countries, such as in Brazil. The dubbing actor who voiced Dom Kolb in the German version was actually the same person who usually dubbed Leonardo DiCaprio.

Director and writer David Zucker said in an interview that the movie's release actually had a three-year wait because the studio tried to convince him to return to direct. Finally, the studio and Zucker agreed that Zucker returns, but as a writer. Then the studio hired Malcolm D. Lee to direct after Lee helmed Soul Men (2008) for the same studio. Before Zucker was added to write, the initial script was delivered by writers Steve Leff, John Aboud and Michael Colton. Zucker rewrote it first together with Lee and later again with his long-time writing partner Pat Proft.

This was the only film in the entire "Scary Movie" franchise that did not feature any main characters from the first 4 films, i.e., Cindy Campbell (Anna Faris) and Brenda Meeks (Regina Hall). In a podcast, Anna Faris said that she was "too old" to return and Regina Hall became "expensive". Initially in 2009, The Weinstein Company decided this was going to be a reboot. Which meant there would be no returning characters.

Although a parody, Jerry O'Connell is the first actor to play Christian Grey of the Fifty Shades of Grey novels since the film adaptation was not in production at the time.

Evil Dead (2013) remake is spoofed in this movie, although it was released only one week before. The "Scary Movie" producers approached the Evil Dead (2013) script and basically spoofed the trailer. The Fifty Shades of Grey (2015) movie was not even in production yet, but it was already spoofed in this film.
